Do you mean putting a design on top of a FSL one? No, it does not need to be fsl, but it will need a nice firm base. So, if the FSL design is sparse, I wouldn't do it.If it is nice and firm, or a bit denser, then yes, you should have no problems.
I stitched these pansies over the top of a FSL bookmark, which I digitized, and had no problems, The pansies are done in satin stitch.

Thankyou you have been most helpful,i am making bookmarks for family children,would like to place their names on them,so they are not lost at school.
I am actually working on some FSL Names now, which are great for bookmarks.I haven't got a great deal of names, but I will add more soon. I also include The initials in 2 sizes in the design packs, great for keys, purses, pencil cases, etc.
